Time to read a book or a catalog. The column can be made by using CNBr activated Sepharose (See Sigma catalog). Most protocols for affinity column manufacture andthe subsequent purificaiton can be found on the web.

Hi, I would like to identify an antigen recognized by an antibody.I was thinking about making a column with the antibody, use celllysates from the cell line that has been shown to have highexpression of the antigen, then run a 2d-gel and sequence theprotein. Any suggestions on protocols on how to make thecolumn, treat the cell lysates and other comments? Thanks,Menno de Winther |
