I have just started culturing THP-1 cells.
After sub-culturing, I wanted to freeze down some of these cells. I froze a vial of cells with my normal freezing media (2x, 20% DMSO and 80% FBS). When I thawed the cells they were viable. After 48h the cells had switched from being in suspension to becoming adherent. Is this from the DMSO? Should I use glycerol to cryopreserve instead? |

Yes I have found that DMSO causes my cells to differentiate. 20% DMSO is very high - 10% is enough. I now use a special freezing medium for my cells as i had a huge problem with differentiation. When you thaw your cells you should spin them down at a low speed and remove the freezing medium to remove the DMSO. |
