I am trying to culture macrophages from whole blood. What I am looking for is a way to assess the purity of my macrophage population adhered to the culture dish.

Use Mac-1 and GR-1(granulocytes) antibodies and do FACS analysis or something like that.

Alternatively, have you ever tried to generate macrophages from monocytes in the presence of GM-CSF ?

Can also try diI labeled acetylated LDL to test their phagocytic activity.

Time of adheration and washing of adhered monocytes are very important. Standardized it.
