I am a Post-Doc at Cornell University and would like some advice regarding the external solution used to patch clamp rat cortical neurons. I am isolating embrionic cells and have seen in the literature that I need to add ATP and glucose to the external solution. I have also heard that I need to prepare the ATP on itself, separate in aliquots, freeze and use it on the day of the experiment. Aparently, glucose should be weighted and added to the external solution on the day of the experiment. Are all these information right? Is there any other detail that I should pay attention about ATP and glucose?
