Question about gel purification of PCR product - (Sep/25/2005 )

hi everyone, i just want to ask the most inexpensive but effective way of purifying a dna band for sequencing. Upon splicing the target band from the gel, where must i store it and at how much volume? Can i store it indefinetely or should i purify the dna immediately?

I just want to ask if its okay to store the 495 bp and 245 bp pcr pruduct I excised from agarose gel in Tris-EDTA since I have no purication kit yet?

What kit can you recommend and if I am going to do it manually, what procedure can I follow? Thank you.

Hello,

I usually store the gel slice in 4C without adding any buffer. A few days is OK.

Check this page for many different methods of gel purification without using a kit http://www.protocol-online.org/prot/Molecu..._Gel/index.html
