    I too fell the same problem with 786-O (renel cell carcinime) It is not responding properly by MTT assay.

    May be in this kind of cell line with over spread we can go SRB ( sulphorhodamineB) is betterly working.

    i tried this assay with cancer cells (MDAMB231 & PC3), and tried the DMSO to dissolve the formazan, but the cellular debris still remains in the well and i think this causes interference in OD readings. how do you rid of that? or fully lyse the cells along with dissolving the DNA? thanks.
