Restriction Enzyme BstBI - (Sep/29/2005 )

I Have to do a digestion with the enzyme BstBI. This enzyme needs a 65°C Incubation to work properly.
--> What do you think, is it a problem to let ite DNA digest at 65°C overnight? degradation?
--> Did anybody work with this enzyme before? What are your experiences with it? Reliable - Nasty?

I don't see any problem (although I never worked withthe enzyme) just don't forget to do a control tube with out the enzyme cool.gif

i get better results without overnight digest. Therefore i use a pcr program : 65°C for 4h, followed by "hold 4°"
