gel extraction of pcr products - (Sep/05/2005 )

Hi,

I want to gel purify my PCR product. for this purpose, is loading 20 ul PCR product on gel sufficient or is it better to load more, for example 50 ul ?

It depends on how much DNA you will need for downstream application. You can load 50 ul or as much as the gel well can hold, or you can load into multiple wells and then combine the gel slices for purification.

I've tried gel purifying 20 ul PCR products before and I've managed to get back approximately 15 ul of DNA at the concentration of 6ng/ul. So if you don't need much for subsequent experiments, 20 ul would be sufficient.
