bacterial culture growth on plate but not in liquid - (Aug/24/2005 )

has anyone ever had a situation where transformed colonies grow on amp plates but not in liquid culture containing the same concentration of amp? i'm pretty sure these are transformed colonies because they grow blue on AMp plates-- XL-1 blue competent cells...

Did u use the same incubation temp?
How about shaking? Did u grow them in a shaking incubator?

Have you checked the pH of the broth? Have you made sure that your innoculation loop was cooled down sufficientlyduring innocultion?....
