real-time thresholds - purpose? - (Aug/23/2005 )
What is the purpose of the threshold on df/dt vs temperature graphs for melting curve analysis?
I have samples that produce the right peaks but underneath the threshold - the computer programme will allow me to move the threshold to include the peaks in analysis - so hence what is the point in it?
Some text appears to say you should discard all peaks below threshold - but if they are so obviously right?????
Any help would be very gratefully recieved

Incase anyones interested - the thresholds for melting curves whenlooking for the presence of a genotype and not doing any quantitation - it is acceptable to lower the theshold to below zero in order to analyse peaks.
