Reuse TAE buffer? - (Aug/22/2005 )

Hello,
Do I always have to use new TAE everytime I want to run my electrophoresis for PCR product?

No, that is not necessary. In my lab we reuse TAE with good results. We change it when it becomes to blue due to the loading dye =)

From some books, they say that new TAE should be used after 1 to 2 times. But I reuse TAE serveral times and I got good results.

I read that TAE is good for 6 hours run, but in our lab we use it for a very long time, and it usually works, so i think you can reuse it.

I reuse it several times. Assuming you are running your gel at 100 volts, it should read approx. 100mAmps. When it goes up to 150mAmps, that is when it is time to change the buffer.

I also use it several times before changing and i have no problems. i only use new buffer when i need to cut the gel to get the DNA i was running...

Resuing buffer is absolutely fine. Depending on your gel box, if you have deep wells where the anode and cathode are, you can cause those areas to turn basic and acidic slightly with repeated use. I mix the buffer up before use and add new buffer to maintain good coverage of the gel.
