Validity of sequences via TA cloning - (Aug/20/2005 )

Hi everybody,We use InstTA clone (Fermentas) to clone 300bp PCR product. As you know Taq DNA polymerase has not proofreading activity. So if we use the cloning kit for sequencing of the product , Are sequences valid? Additionaly I know we can sequence the product directly.

It depends on your downstream analysis.
If you would like to have protein expression, proofreading enzyme is recommended.
If you just like to identify what the gene is, Taq should be fine.
