Is it possible to gel purify the ligated product? - Avoid non cut vector or self-liagted vector contam (Aug/13/2005 )

Hi all!

I am thinking of ways to avoid non cut vector or self-liagted vector contaminating the transformants, anyone try gel purification before?
what condition is better?

I don't think it really is, most ligation reactions give a whole lot of different product on a gel, so it would be hard to separate the right product. Also, in a ligation reaction you're most likely not going to use a lot of DNA, so you will not have much left after purifying.

Hi vairus,

Maybe you're right,
I tried to separate liagted products by gel elecrtophoresis,
then what i can only found is large size smear(beyond 10kb)
on the gel and i have no idea about what is that! blink.gif
