Amount of genomic DNA for PCRamplication - (Jul/24/2001 )

Can you tell me usually how much genomic DNA should be used in a 50ul PCR reaction using Taq polymerase? The DNA was obtained from a feline cell line.

For example,with dna mouse (tail), I soak the tip pipet in dna tube and I soak in pcr mix.I have make pcr with this protocole and it's ok, as with 1 microL,2 microL but no pcr product with 3 microlThe DNA was estimated at 260nm and it was at 400ng/microL so, the amount must be about 500ng/microL.The genomic DNA must be absolutly clean for pcr testgood luke
