FLAG-Immunoprecipitation - (Aug/10/2005 )

Hi everyone:

I am using the EZview Red ANTI_FLAG M2 Affinity gel for IP of flag tagged proteins.

My query is, is it necessary to do a protein assay before loading the lysates with Affinity gel? If yes, preferred method for protein assay.

No, do the assay after eluting the protein from the beads, otherwise you are measuring total cellular protein rather than the amount of protein you have pulled-down.
