Suggestions for genomic library screening ! - Design of probe (Aug/02/2005 )

Hi

I am doing a genomic library screening of raabit.The gene has been sequenced from human,mouse and rat.It would be great if someone could suggest me what should be the ideal probe to fishout the gene from rabbit genomic library.Is it a good idea to design an oligo based on the homology sequence alignment of human,mouse and rat?

Yes. Try and use a region that is conserved across all three species. If you can't find a region that is 100% conserved then bias towards rat/mouse rather than human. Remember to check that the probe you design doesn't cross hybridize to any other genes in the known genomes.

If you are going to use the cDNA approach (I think using a oligonucleotide probe would be simpler) then either rat or mouse would be equally good.
