Re-transforming-- Is re-sequencing necessary? - (Jul/09/2005 )

I have a construct that is sequenced and in the form of a glycerol stock. I'm having problems growing the glycerol stock for maxi preps. I believe something is wrong with the glycerol stock.

I am thinking of re-transforming the construct into DH5-alpha, plating and then making a new stock.

My question: If I were to re-tranform, then plate with a selective antibiotic, then pick a colony and grow it up, would I need to re-sequence the construct?
