A quick question about sonication to lyse cells - (Jul/05/2005 )

Hi everybody,


I just need to confirm one thing for sonication :
I use it to lyse some mammalian cell lines for
protein extraction and purification. Just wondering if
also break the nucleus as well? I presume it does!

is there anyway to set up the sonication apparatus so that the nuclei are not broken open? ie by changing the frequency, probe size/shape, duration etc

I seriously doubt it. Any sonication with enough power to break the cell wall is also going to get the nucleus.
