Should the PCR effiencies of the standard curves be the same as the unknown samples in absolute quantitation and in the relative quantitation based on standard curves?

Yes, otherwise your quantitation in not vaild.

It would be quite messy and complicated since the PCR effiencies have to be the same. Should they have identical PCR effiencies or is that a range of values where they can be in comparing to each other? What kind of quantitation methods do you use for making it easy?

I am wandering how you could measuring you PCR efficiency of your unknown samples.
As I know, as long as you are using the same primer for both of your standard and unknown samples, the PCR efficiency should be the same.
