how to thaw intestinal samples? - (Jul/01/2005 )

Hi all laugh.gif

I have some intestinal sample that i kept in -80oC and i would like to extract DNA from the sample. The quantity of the DNA is important for my study because i am going to quantify the amout of DNA in the intestinal sample. What is the best way to thraw the sample to minimize the lost of the DNA? Should i thraw it in room temp or 4oC??

Thaw on ice...better yet, why thaw at all. Pull it out of the -80, break off a chunk of gut, put it in the DNA extraction buffer, put the rest of the gut back in the -80.
