does anyone have experience of doing phagocytosis - (Jun/23/2005 )

i am trying to induce phagpcytosis using opsinized latex beads to see some tyrosine phosphorylation? can anyone give me some advice as the tme point and procedure in inducing phagocytosis?

what type of cells?... macrophages will take just about anything up rather quickly, (within an hour), I would try a time course and see what works best for your cells. lets hear some more info so we can help you more. smile.gif

tyrosine phosphorylation is a very quick phenomenum (5-30 minutes)

Both Macs and DCs phagocytosis very quickly-within about 5 mins (especially since you have coated them). I would do 2 things one, synchonize your experiments. We do this in our lab by turning down the centrifuge to below 15C for 5 minutes and then putting them in the incubator. Time zero being the time you take it out of the centrifuge. As for Phosphorylation..I also look at phosphorylation which is downstream of Tyrosine Phosphorylation and we can see that as quickly as 10 minutes so probably a time course of 5,10, 15, and 30 minutes is best. Hope this helps.
