I also use the Qiagen kit among others (Edge Biosystems, Sigma, Promega, etc). I try to only use them for important downstream applications, i.e. sequencing as alkaline lysis is just fine for checking for the basics.

It is valuable to know the chemistry behind the protocols and be able to do them kit-less. But I have to say, time is money too and I wouldn't be able to do three projects at a time without kits and pre-cast gels. smile.gif

Pfffft. I always cast my own gels. Takes 10-15 min tops to assemble and cast. That way I get a stacking gel too.
