Transfection of 293T cells on coverslips - (Jun/16/2005 )

I recently attempted to transfect 293T cells grown in 6 well plates on round coverslips. As I was in the process of washing my coverslips prior to probing them with antibody, I found that most of the cells were sloughing off. I had been fairly careful about not disturbing the cells too much since they come off plates very easily, but I still had this problem. Does anyone have any tricks for helping 293T cells stick to glass coverslips? Thanks!

Try Treating the coverslips with poly-L-Lysine before plating. There are also other products you can use, Like Extracellular Matrix Gel (both can be obtained from sigma).
