help on transformation - BAC clones transformation (Jun/11/2005 )

Hi,

This is my first time working with BAC clones. I have tried using heat-shock method to transform BAC into DH5a competent cells. But it doesn't work!

I read that electroporation is the usual preferred method for transformation of BAC clone, but is there other methods around that i can use?

My friend worked on YAC and BAC and she said transformation rate into E.coli cells was quite low.

I asked her just to prolong the on-ice incubation time upto 1hour, she did and got a better result, you may try it!

Do u mean, the on ice incubation before heat shock or after heat shock at 42 degrees?
