How to get the sequence detected in Northern blot? - (Jun/08/2005 )

hi, there,

With part of my gene as probe, I get two band in the northern blot, I want to know their sequence, but I don't have idea how to do it. Could some one here give me some suggestion? Thank you in advance!

Both bands should have a segment of sequence that is complementary to your probe (because it anneals to them), so their sequence is the gene sequence itself. The only difference between these two RNA transcripts (two bands) is where they start and that can be checked by doing primer extention assay using the same probe u have.
