separation of nucleus protein - lysis buffer (Jun/02/2005 )

My lysis buffer is:
NaCl 100 mM
Tris HCl 20 mM (ph &,4)
EDTA 2,5 mM
Triton 1%
SDS 0,1 %
after 15000*g,20min, i get supernatant,is the supernatant include the whole nucleus protein?

if we assume cytoplasm is lysed before nucleus, i would say you'll get both nuclear and cytoplasmic proteins...


for separation of nucleus proteins, i use the protocol described here.
