Which RT primer is better for qPCR - (May/29/2005 )

Hi all,

I'm going to be doing qPCR and want to know which type of primer is best for the RT part (random hexamers, 3' reverse, oligo dT)?

Hi molgen,

I guess which method to use in your RT depends on what you are trying to quantitate.

If it is just the mRNA message then oligo dT should suffice, however if you would like to quantitate all transcripts from a particular gene (EST's and splice variants) then it would be better to use random hexamers because you will then to creating cDNA for all the RNA population.
