circular vector propagation - How? (May/26/2005 )

I have a circular recombinant vector and I have been told that I can propagate it and never have to re-buy it. Do I do this by transforming the vector into DH5 alpha cells and then growing colonies and freezing the pellets? If so this means that every time I want to ligate something new into the vector I have to purify it from the bacteria right?

I am rather new to this and would really appreciate the help.

You can keep a stock of the transformed bacteria with your vector and also store the purified DNA at -20. When you run out of vector, you can always grow more.
