PCR on plasmids? - (May/20/2005 )

Is it possible to do PCR on plasmids, or does the plasmid have to be cut linear first?

If it's possible, how does extension of the sequence stop at det correct position when the DNA is circular?

no the plasmid does not have to but cut for PCR.

you have a limiting amount of plasmid, the first transcript products from the reaction will carry on past the primers, but as the reaction progresses, it is the product determined by the primer pairs that dominate and that is what you see on a gel......all the other products are there in the reaction mix but not abundant enough to be seen and accounted for.
