maxiprep - bad concentration of DNA (May/18/2005 )

hello all,

I have problems with the concentration of DNA after maxiprep, because I always obtein a viscous solution with something like precipitate but I donĀ“t know if this is DNA or rubish.

how can I obtein a good concentration?
Is it problem with the quiagen column?

Hi,

Are you washing your coulumns properly? Maybe your eluate is contaminated? I dont understand what you mean when you say that you obtain a viscous solution...do you mean when you collect your eluate prior to centrifugation?

if the solution is very viscous it is usually a sign of very high concentration of unsheared DNA in solution, however with a precipitate......what color is this precipitate and does it dissolve upon heating?
