multiplex PCR - template and primer concentration (May/15/2005 )

In a 25 micro lit. Multiplex reaction, i'm using 5 micro lit. of DNA mix (10ng of template DNA 4 different subtypes) of the same bug and 1 micro lit. of each forward and rev.primer from primer mix ( each primer at 0.5 micro Mole from 4 different subtypes). Haven't been a successful Multiplex,
