Mutagenesis - How to use quick change method - (Mar/23/2002 )

I want to use quick change kit to make site-directed mutagenesis. But becuae I am in a small protein crystallography lab, my boss told me that we can't afford the expensive kit. So I want to know where can I find the articles describing this method? Or can we use ordinary Taq polymerase instead to carry on my work? Any beneficial suggestions will be very appreciated.

There are 7 references at the end of the protocol for quick change. Try Stratagene's web site for the protocol. It would be a pain to list them here. If you can avoid it use a thermostable polymerase that has 3'-5' exo activity. Try to avoid taq. Perhaps you could talk a company into giving you  a free sample.
