Hi,

Can I use standard curves generated from brain RNA to quantify my patient's RNA and other normal RNAs that are isolated from cell culture???

Good question. I seen a lot of literature where standard curves were generated from rna using cell cultures and then used to establish quantitative values for specimen samples derived from tissue or blood. If this is exceptable then the inverse should be too.

However, if you were able to quantify the brain RNA why not use that same method to quantify the cell culture RNA.

Patient cell culture is from whole blood ( I can not get brain tissue from my patient). Although, my target gene expression is too low in blood, its expression is high in brain. When I tried to generate standard curve using whole blood cell culture RNA dilutions, there is not any amplification with low dilution series. I thougth, if I can use brain RNAs for dilution, probably, I will solve that problem!!!!!
