Melting curves in RT-qPCR-help! - (Apr/23/2005 )

How do i know how many cycles should i run the melting curve and at what temperatures and for how long for a spesific product?


Besides i have seen some examples of melting curve programs and it says "ramp". What does it mean with ramp? For example this one;

1 Cycle (Dissociation Curve):
95°C for 15 sec
60°C for 20 sec
95°C for 15 sec [Ramp 20 min].

Dear seasons,
Are you sure is [Ramp 20 min], beside that you only have 15 sec ?
Could you please tell me what machine are you using?

According to the program it is 60°C for 20 sec and 30sec with 95°C. do you mean that the ramp should be 50sec and not 20min? unsure.gif

It is a light cycler.
