In DNA pool sequencing to caculate the allele frequence of a SNP, How many samples should I use and How can I make the DNA quality be equal?

Make sure all your DNA is equimolar by using a spectrophotomer then add the same ul amount of each for your pool. There is no fixed number that should be in your pool - less (ie 5) increases the odds of finding rare alleles while more allows you to scrren greater numbers of samples with the chances of missing a rarer allele here and there.
