restriction enzime cutting of genomic DNA - (Apr/06/2005 )

How many units of restriction enzyme per microgram of genomic DNA do you use? and How many hours do you digest it? thanks.

Hi Marvella,

you should check the pack insert of the restriction enzyme you are using, they give the number of units of enzyme in terms of amount of lambda(plasmid) DNA digested in one hour. If you are digesting genomic DNA( which is more complex) of course you would add more units to the reaction.

NEB have a really good site about restriction enzymes, but it's best to goto the website from which you brought the enzyme from and download the pack insert!
