immunohistochemistry - tissue won't stay on slides - (Apr/05/2005 )

Hi,

I have some brain slices that have been mounted onto slides and the tissue will not stay on during immunohistochemistry. I have tried to dry the slides overnight (>15 hours) and the sections are still comming off. I am worried that additional drying may cause antigen degradation. Whenever I have had this problem before, a few hours of drying has been sufficient. Any advice on how to get the tissue to permanantly stick to the slides would be greatly appreciated.

Try using poly-lisyn on slides before mounting of tissue

Try Bond-Rite Slides from VWR (cat#16002-536).
They work great and you can get a case for around $200.
With these slides you do not have to bake them AT ALL!
I use them for in situ's and immunos and they are wonderful.

I agree, buy the slides from VWR or Fisher that are electrostatically treated to give good tissue sticking. Well worth the modest extra cost. WE use them standard for frozen sections.

Vector labs also has a treatment solution that you can use if you need more heavier washing and temperature treatments like for in situ.

For routine stuff I find PLUS charged slides from VWR (Superfrost Plus) to be great. For stuff like brain and bone which likes to come off I like the silane coated slides from Sigma.

Fisher PLUS slides are the best. We boil our sections for 30 min in a microwave and they do not come off at all.

Try these slides.
