Stable Transfections - do you NEED to linearise the plasmid? (Mar/31/2005 )

hi all

this may be a silly question, but i was wondering whether you need to linearise your plasmids before transfecting them into eukaryotic cells for stable transfection. i've heard arguements for and against. what do you guys do?

there was a discussion here about linearize plasmid before transfection. I think this is not a necessity due to the fact you further select cells that are ok. Moreover, linearize a plasmid do not prevents from an other cut in the DNA in the cell before integration...
