siRNA transfection into 3T3-L1 - (Mar/31/2005 )

Hi everyone,
I have done siRNA transfection into 3T3-L1 cell lines (pre-adipocytes) several times with siRNA made from our Lab and RNAiFect Transfection Reagent from Qiacen. However, I found that the cells couldn't differentiated well after transfected (over 90% non-treated cells got fully differentiation compared with 60-70% in the case of transfected cell).

Is there anyone has done the same my experiment now. Could you help me how to solve the problem?

First, do you have a control to rule out the effect of your transfection reagent?
e.g. is this effect due to transfection reagent you used??

have you tried another transfection agent?

why do you select this cell?
why do not use NIH 3T3 cell?
