Good buffer needed! - (Feb/17/2005 )

Hey all!

Small question for protein people. I want to use a nontoxic buffer with pH 5-6 to my proteins, which buffer you recommend?
Could just PBS be used or something?

Depends on what you want to do... adjusted PBS or HBSS would seem fine to me... or some Tris based buffer.

I frankly don't know!?!? The more acidic Tris solution we use is 6.8...

tris cannot be used at ph less than 7.5. its buffering capacity diminishes below that pH. It wont buffer at all in the range 5-6. The buffer capacity of can system depend on the pka of the compounds used. for tris it is 8.3

PBS is fine I guess

A good choice for pH 5-6 is citrate-phospahte buffer

If it is 5, i suggest acetate buffer
If it is 6, i suggest phosphate buffer
