Nuclear RNA knockdown with siRNA - is it possible? (Feb/09/2005 )

Hello,

I'm trying to knockdown the amount of a nuclear RNA. Does the Dicer-mediated degradation of the targeted RNA occur in the nucleus or in the cytoplasm??

If it occurs in the cytoplasm, does polyadenylated nuclear RNA get exported to the cytoplasm before it is localized to the nucleas??

Is knocking down a nuclear RNA feasible?

There is a very recent paper reporting RNAi works in the nucleus. The abstract is here

my experience is that in the nucleus, antisense works a lot better than RNAi.

For mammalian cells, try antisense oligo to knock down nuclear-retained RNA.
