Met trouble in linking DNA!!!! - (Nov/21/2001 )

My insertion have 5' Sac1 and 3' Xho1 digestion site, the target plasmid has only Bgl2 site, how to link them? where can I find the protocol? Thanks a lot!

You have two choises, either use PCR with primers containing the Bgl2 site, alternatively fill out the DNA ends of Vector as well as insert with the Klenow fragment, and make blund end ligation

Why not just PCR with primers with those restriction sites? In other words, create those restrictions sites in your insert.
