Where do I start? - (Jan/19/2005 )

I have determined that a 3.5kb plasmid is linked to the production/immunity of my peptide of interest. How do I go about sequencing/cloning etc the plasmid? Any suggestions? Need rapid results. Thank you.

I assume you are talking about a natural plasmid...

i think you first need to obtain the plasmid pure - if it is the only plasmid then no problem, otherwise you may have to run your plasmids on gel and cut it out. then i would try restriction with several common enzymes, once you manage to cut it even once, you can clone into a commercial vector (PUC etc.) which you have cut with the some enzyme. then sequence from vector primers.
