RNA Isolation - (Jan/13/2005 )

I met with one phenomenon during isolating bacteria RNA. After adding the isopropyl alcohol, some fibrils-like material immediately appeared. In fact, the pellet of RNA should not be seen in such a very short time period. Anyone out there can tell me what happened, and your suggests will be greatly appreciated.

Though I have not seen that fibrill like structure in your case but I suppose there is contamination of genomic DNA because it is genomic DNA which forms whitish fibrill like structure immediately after addition of isopropyl alcohol.So you can try some other protocol also for RNA isolation.

agree with promilasharma.adjust the PH value of your extraction solution.

I thought fibril- like material of DNA contamination firstly, but the later gel-running showed two clear band , 23srRNA and 16srRNA, being no any other bands. In the other hand, isolating the RNA from the control cell with the same solution and procedure did not produce the same results.
