DGGE and reamplification - (Dec/29/2004 )

Hi,

I am trying to reamplify PCR products extracted from DGGE gel slices but with no success. The extraction method I am using is incubating the gel slices containing the DNA in 50 microl. DDW overnight at 4C. I would very much appreciate any help you offer regarding this problem.

We use the same overnight incubation in ddw but the next day we vortex the tube and prior to PCR stir the slice around in the water with a tip this works for us alternatively you could try freeze thaw treatment
