problem with blocking - Blocking endogenous activity (Dec/22/2004 )

Hello. I am working with frozen sections and experiencing problems with high background.
Can I block for endogenous peroxidase (these samples contain blood) with 0.3% H202 in sodium azide and still visualize with BCIP/NBT?

I have only blocked in normal serum made to the source of the primary, but I've read that you can block in normal serum from the species of the sample. Has anyone tried this? Can you explain to me how it would work?

Any other suggestions would be greatly appreciated.

Try blocking with 5% serum from the source of your secondary antibody. This usually cuts down background
