Binding of protein to immunoassay plates - binding in denaturing conditions (Dec/14/2004 )

Hi,


Does anybody knows wether it is possible to coat protein on immunoassay plates(NUNC maxisorb) under denaturing conditions (typically few M urea or few M guanidine HCL)? I have a big doubt about that.

usually you don't have harsh denaturating condition while coating, since you're dilluting the protein solution in coating buffer
I successfully coated protein denatured in 8M urea to medium-binding-capacity plates as well as to maxisorp plates. but I used only 2µl of protein solution with 98µl coating buffer, so the actual urea concentration was only 160mM while coating. worked fine in my hands.
