Separation of human DNA from virus DNA - (Nov/17/2004 )

HI!

I would like to separate my virus DNA from human DNA. The virus is 8kb and circular. I´ve been thinking about magnetic beads. Maybe you can attach a certain sequence to the beads so the human DNA could hybridize? I don´t know the sequence of the virus. I´m open to everything. I hope someone has a suggestion to me smile.gif

8 Kb and circular sounds like it's ideal for a miniprep to me - depends how much DNA you can get. You could try a qiagen miniprep or something?
You could also try a caesium chloride gradient. This will separate your viral dna as a discrete band in the centrifuge tube, from the much larger genomic DNA.

also, check the roche catalogue - they have a viral dna purification kit.

Thank you for your suggestions. I think I don´t have enough virus to use miniprep. One in my resarch group did it and it didn´t work for her and our samples are similar. I also think you need quite a lot of material if you are going to use the cesium chloride cetrifugation.
I´m giong to look at roche´s purification kit.
