Mitochondrial DNA problem - (Jan/12/2009 )
Hello alll
could you please tell me if there a problem to use whole genomic DNA in order to do a PCR reaction for a mitochondrial gene
as the pcr reaction dont give any bands on the gel
should i isolate mitochondrial DNA at first????????

I use the Puregene protocol from Qiagen (formerly owned by Gentra Systems) and it works well for getting both nucleic and mitochondrial DNA. I amplify the HVR1 and HVR2 genes form the d-loop and those are actually some of my most successful reactions and I just use 1ul of DNA from the Puregene protocol.
