who has the detail triparental protocol? - (Jan/12/2009 )

as the topic, who can help me? thank you very much.

Not enough information -- there are many triparental protocols, depending on what species or strains you're mating.

In general, you need a strain with a helper plasmid capable of mobilizing other plasmids, the donor strain with your cloned DNA in a mobilizable vector, and the recipient strain in which the mobilizable plasmid can or can't replicate, depending on what you're doing. The strains usually need differing antibiotic sensitivities so you can select against the helper strain, the donor strain, and the cells of the recipient strain that didn't receive the mobilizable plasmid.
